How To Extract Nucleic Acid By Magnetic Bead Method

For first-time users of magnetic bead nucleic acid extraction kits, paying close attention to the key details of handling the magnetic beads, in addition to other routine procedures, will greatly increase the success rate.

1. Pre-Treatment of Magnetic Beads

  • Magnetic beads will settle when left standing. Before use, they must be fully resuspended by vortexing or vigorous pipetting until a uniform suspension is achieved. This ensures consistent volume measurement for each use.

2. Key Operations During the Experiment: Details Make the Difference

2.1 Magnetic Bead Binding Time

  •  After adding the magnetic beads, ensure sufficient room temperature incubation time (typically 5-10 minutes). Gently mix the solution several times during this period to allow thorough binding between the nucleic acids and the magnetic beads.

2.2 Magnetic Separation Technique

  • After placing the centrifuge tube on the magnetic stand, allow enough time (usually 2-5 minutes; magnetic beads from Shanghai Lingjun Biotechnology require only 10-30 seconds due to their strong magnetic properties) for the solution to become completely clear and for all the magnetic beads to be fully adsorbed to one side of the tube. Do not rush to aspirate the supernatant immediately, as this may draw incompletely adsorbed beads and lead to nucleic acid loss.
  • When aspirating the supernatant, avoid touching the aggregated magnetic beads with the pipette tip. It is better to leave a small amount of liquid behind than to disturb the beads.

2.3 Adequate Drying

  •    After the final wash, open the tube cap and let the magnetic beads air-dry at room temperature on the magnetic stand for 2-5 minutes. This allows the residual ethanol to evaporate completely.
  •    Key indicator: The beads are sufficiently dry when their surface changes from a smooth, glossy brown to a matte, non-reflective texture. Avoid over-drying, as this will make nucleic acid elution difficult and significantly reduce yield.
