From Hot Spring to DNA Identification
In the summer of 1966, Hudson Fritz, an undergraduate at Indiana University, moved into a cabin on the edge of Yellowstone National Park. He was then working for microbiologist Thomas Brock.
Brooke was convinced that certain microorganisms could survive in temperatures far exceeding expectations. Consequently, Fritz made daily trips to the hot spring area to collect bacterial samples. On September 19 of that year, he successfully cultured a pale yellow microbial sample from the Mushroom Hot Spring. Under the microscope, he observed abundant cells collected from water near boiling point.

“I saw something my predecessors had never seen,” recalls Fritz, now at the Sandford Burnham Prebises Institute for Medical Discovery in California. “To this day, the sight of that microscope image still sends shivers down my spine.”
Three years later, Fritz and Brock described one of these bacteria, naming it “Thermophilus aquaticus.” This bacterium thrives optimally at 70°C. In 1976, a team from the University of Cincinnati, Ohio, isolated an enzyme from Thermophilus aquaticus—a “DNA polymerase” capable of synthesizing new deoxyribonucleic acid (DNA) at 80°C.
Seven years later, a substance dubbed “Taq polymerase” proved ideal for biochemist Kary Mullis to develop the polymerase chain reaction (PCR) technique, which can rapidly amplify a single DNA fragment into thousands of copies. Since Mullis required high temperatures to denature DNA strands, he needed a polymerase capable of functioning at elevated temperatures.
Today, PCR technology has become an indispensable tool, widely applied across multiple fields. In the medical field, it is used for organ transplant matching and cancer diagnosis; in criminal investigation, it enables DNA fingerprint identification.
