To avoid the risks associated with organic solvents, traditional extraction reagents rely on ethanol or isopropanol as nucleic acid stabilizers. However, these alcohols are flammable, explosive, and volatile, requiring special conditions for transportation and storage (such as low temperatures and explosion-proof packaging). In contrast, alcohol-free reagents eliminate these safety hazards through alternative formulations (such as polyethylene glycol and oil phases), reducing transportation costs and enhancing the stability of the reagent after opening.

Improving operational convenience and compatibility with automation, alcohol-containing reagents require manual intervention for addition in automated processes, whereas alcohol-free reagents can be preloaded directly into the kit, enabling fully enclosed automated operations.
